Description:
Our research interest is focused on gene discovery, functional gene annotation and signalling in cancer. We are developing genetic tools for cell-based screening as well as technologies for high-throughput forward and reverse genetics in mice. Examples include transposon platforms for insertional mutagenesis in mice. They allow genome-wide screening for cancer drivers that are difficult to identify with other methods (e.g. sequencing) and currently create comprehensive catalogues of cancer genes for various solid and haematologic neoplasms. We also utilize these systems to uncover mechanisms underlying tumour evolution, metastatic spread and treatment resistance (e.g. Science. 2010 Nov 19;330(6007):1104; Nature protoc. 2017 Feb;12(2):289).
In hypothesis-driven approaches we are applying advanced reverse genetics in mice to generate specific models of digestive tract cancers and their subentities. We deploy these models to study molecular pathogenesis and signalling in cancer, as well as for preclinical research to develop novel diagnostic and therapeutic strategies (e.g. Nature. 2018 Feb 1;554(7690):62).
Another focus is the functional annotation of cancer genomes. To this end we develop CRISPR / Cas9-based approaches for somatic genome engineering in the mouse and use them for targeted mutagenesis, genetic screening and chromosome engineering in the context of gastrointestinal or haematopoietic cancers (e.g.PNAS 2015, Nov 10;112(45):13982).
We are an enthusiastic team and offer intensive training and mentoring. We use a broad spectrum of scientific approaches and techniques, including functional genomics, genome informatics, biochemical approaches, in vivo molecular imaging and others. The proximity to the clinical facilities allows insights into clinically relevant aspects of cancer. The successful candidate will be employed under the umbrella of the German Cancer Consortium (DKTK) at DKFZ as well as at the newly founded Institute for Molecular Oncology and Functional Genomics (Technical University of Munich). The Institute is located at the newly built Center for Translational Cancer Research, TranslaTUM, in the heart of Munich.
