Project background Colorectal cancer develops in a multi-step-process from normal epithelium, through a pre-malignant lesion (adenoma), into a malignant lesion (adenocarcinoma). A minority of about 5% of adenomas will ultimately progress into cancer. So far, solid tools to predict which adenomas are at high risk of progressing to carcinoma are lacking. We have previously identified DNA copy number changes, RNA transcripts and proteins associated with adenoma to carcinoma progression. This includes AURKA, a protein kinase, and TPX2 at chromosome 20q as major candidates driving colorectal adenoma-to-carcinoma progression. The ultimate phenotype induced by genomic alterations is due to changes in expression and activation state of proteins, e.g. phosphorylation. This translational research project will yield basic knowledge on colorectal cancer development and focuses on protein phosphorylation and functional validation of candidate driver kinases. Project The prospective candidate will perform the experimental work and data analysis aimed at identifying (phospho)-protein signatures involved in malignant transformation of colorectal adenomas. The candidate will also functionally validate the role of candidate proteins in organoid models of colorectal adenoma to carcinoma progression. Moreover, he/she will analyse the functional effects in vivo by injecting manipulated cells in nude mice.

Benefits

The temporary employment will be for a period of 4 years in an exciting and actual topic of cancer research embedded in an enthusiastic research group that is internationally oriented and performs multidisciplinary research. The gross salary per month will be € 2.529,- to € 3.115,- according to the PhD (OIO) scales or € 3264,- to € 4177,- according to the postdoc researcher function group 55-60, and depends on previous experience.
