A postdoctoral research fellow position is available in the Hagan laboratory, University of Texas Health Science Center at Houston. Our research group focuses on microRNAs and their post-transcriptional regulators with an emphasis on developing molecularly targeted cancer therapeutics. Of particular interest is a newly discovered form of post-transcriptional gene regulation involving 3’ RNA uridylation mediated by Terminal Uridylyl Transferases (aka polyU-polymerases). Recent and provocative studies highlight that dysregulated TUTase activity alone and in concert with the onco-fetal LIN28/let-7 pathway are hallmarks of poor prognosis in multiple cancer types.
Specifically, we demonstrated that the TUTase Zcchc11 works with the oncogene and reprogramming factor Lin28A to block biogenesis of the tumor suppressor let-7 microRNA family (NSMB, Cell). Moreover, we demonstrated that ZCCHC11 inhibition caused regression of established LIN28A cancer xenografts. In addition, E2F1 and the TUTase ZCCHC11 are a dual gene classifier for poor breast cancer prognosis, independent of tumor subtype and LIN28 status. Consistent with this finding, many polyadenylated mRNAs are characterized by 3’ RNA uridylation in a gene-specific manner that is implicated in regulating mRNA turnover. Current research on 3’ RNA uridylation is reminiscent of the microRNA field circa 2002. As such, we seek a highly motivated researcher who has a strong commitment to work in an emerging research area with translational implications. Projects rely upon in vitro methodologies, cell culture studies, high-throughput genomics, small molecule drug screens, genome-wide CrispR loss-of-function studies, and in vivo functional analyses in genetically engineered mice with an emphasis on cancer models.
Qualifications:
Interested applicants should be highly motivated and have a Ph.D. degree with a strong background in molecular biology and preferably in mouse genetics. Prior publication in internationally recognized journals is required and candidates should be fluent in spoken and written English.
