POSTDOCTORAL FELLOW – HIV REPLICATION AND THERAPEUTICS

A postdoctoral position is available in the Viral Mutation Section (VMS), HIV Drug Resistance Program (HIV DRP), to perform basic research on HIV replication and therapeutics.

Our goal is to understand how interactions between HIV-1 and the host cell influence its replication and to develop novel therapeutics for the treatment of AIDS.  We are active in three major research areas that are focused on the interactions between host restriction APOBEC3 proteins and viral Vif protein.  First, we have identified several candidate host genes that influence Vif-mediated degradation of APOBEC3G and are characterizing the interactions of the proteins expressed from the genes with APOBEC3G and viral Vif protein.  Second, we have used APOBEC3F to efficiently label viral preintegration complexes and are using this approach to gain insights into the early stage of HIV-1 replication.  Third, we are developing a gene therapy approach for the treatment of AIDS based on delivering Vif-resistant APOBEC3 genes to target cells of infection.  We have also carried out a high-throughput screen to identify small-molecule inhibitors of the Vif-APOBEC3 interactions, and we are characterizing several candidate molecules as potential lead inhibitors of this novel antiviral drug target.  Further details and a list of relevant publications can be found at http://home.ncifcrf.gov/hivdrp/Pathak.html.
