Postdoctoral Long Title: Design and synthesis of ionizable cationic lipids for nucleic acid delivery.

We're currently looking for talented scientists to join our innovative academic-style Postdoc. From our centre in Boston, USA, you'll be in a global pharmaceutical environment, contributing to live projects right from the start. You'll take part in a comprehens ive training programme, including a focus on nucleic acid delivery and development, given access to our existing Postdoctoral research, and encouraged to pursue your own independent research in cutting edge laboratories. It's a newly expanding programme spanning a range of therapeutic areas across a wide range of disciplines.

Pharmaceutical Sciences IMEDis a global organisation that is responsible for engineering delivery systems for therapeutics. This ranges from familiar technologies like pills and solutions, to more advanced preparations for inhalation and nanomedical applications.

This is an exciting opportunity for you to break new ground in a scientific area which is of critical importance to AstraZeneca's future drug portfolio. Nucleic-acid based therapeutics, like messenger RNA and antisense oligonucleotides, are new therapeutic modalities of strategic importance to AstraZeneca. These modalities have the potential to transform the way we treat disease but their successful therapeutic application is currently limited by inefficient intracellular delivery.

You will aim to study the role of excipients for the intracellular delivery of nucleic acid therapeutics. This project will be driven by your expertise and focus on the following areas:

  • Design and synthesize novel delivery systems for nucleic acid therapeutics.
  • Apply medicinal chemistry principles to design, synthesize and formulate lipid and polymeric nanoparticles for mRNA delivery.
  • Perform library screening of novel lipid based nanoparticle formulations for the advanced drug delivery team.
  • Collaborate with biologists to explore new pathways to drive the delivery of therapeutic macromolecules to different tissues

You will be embedded in a multidisciplinary team that will present you with the opportunity to learn f rom considerable internal expertise across exciting technologies including high throughput screening, dynamic combinatorial chemistry, and emerging synthetic technologies for delivery of new chemical modalities to address unmet medical needs. You will also be exposed to experts working on developing cutting-edge drug delivery technologies. This breadth and depth of expertise will support you in producing high impact work that you will publish and present to the external international scientific community.

Essential

  • Ph.D. degree in synthetic organic chemistry or pharmaceutical sciences with experience in nanoparticle design and synthesis.
  • Experience in synthesizing lipids, peptides or polymer derivatives as biomaterials for the drug delivery.
  • Prior experience with lipid nanoparticle formulation for nucleic acid delivery (siRNA, antisense or mRNA etc.) is preferable
  • Experience with analytical techniques for characterizing nanoparticle formulation (DLS, Zetasizer, fluorescent assays, HPLC, GPC etc.,).
  • Proven ability to drive innovative research though internalisation of new techniques and technology, as well as forming internal and external collaborations.
  • Track record of scientific excellence, with a strong publication record and engagement with the scientific community through both conferences and collaborations.

Desirable

  • Strong publication record in the field of organic chemistry, polymer synthesis and nanoparticle formulation.
  • Knowledge of drug delivery technologies for the intracellular delivery of macromolecules.
  • Experience with tech transfer and scale up with production of nanoparticle f ormulation for testing in rodent and higher species in vivo.
  • Ability to lead projects, interact and effectively collaborate in a multidisciplinary environment comprised of chemists, biologists and pharmacologists
