Description

The Department of Cardiovascular Sciences has the entire range of competences from basic science, over pre-clinical modeling to applied clinical and epidemiological research taking a systems approach to the complexity of cardiovascular disorders. The division of Experimental Cardiology is part of the cluster 'Cardiac structure and function' investigating cardiac structural, contractile and electrical remodeling processes during cardiovascular disease both at cellular and organ level including molecular mechanisms, left and right ventricular mechanics and impact of different interventions and anaesthetics. The research will focus on the role of calcium microdomain signaling involving ryanodine and IP3 receptor channels in cardiac myocytes, and the modulation of such local signaling in health and disease (ischemic cardiomyopathy, atrial fibrillation). Previous research has identified local calcium microdomains generated by IP3 receptors in the nuclear envelope, that control transcription, calcium domains generated through ryanodine receptors in control of contraction and contraction-excitation feedback, as well as interactions between ryanodine receptors and IP3R. The specific aims for the project are (1) To address the mechanisms for generation and interpretation of microdomain calcium signaling in excitation-contraction coupling and excitation-transcription coupling. (2) To develop methods for validation of preclinical findings in human cardiac tissue, using direct isolation of cardiac myocytes from biopsies as well as cardiomyocyte differentiation from induced pluripotent stem cells. (3) To develop advanced imaging of local calcium, IP3 and ROS, and novel tools for pre-clinical research using in situ gene manipulation thus enhancing translational potential. The research is supported by Odysseus financing to L. Roderick and Program Financing to K. Sipido ('Heterogeneity in ischemic cardiomyopathy'). The lab has extensive facilities for functional confocal imaging, electrophysiology and access to relevant large animal models. The research group has several postdoctoral and doctoral students. We are recruiting two PhD students who will be assigned to the aims (1) and (2), where one student will focus on how IP3 receptors contribute to microdomain signaling and one student on calcium microdomains in excitation-contraction coupling. A junior postdoctoral scientist will be recruited to develop novel experimental approaches, genetic and imaging tools that they will combine to understand how calcium signals for contraction and transcription are discriminated between.
